摘要: 应用反转录一聚合酶链式反应 (reversetranscription poly merasechainreaction ,RT PCR)方法 ,亚克隆出大鼠小脑和肾脏NOScDNA ,并以此为探针 ,通过Northernblotting分析和定量PCR方法 ,测定了大鼠中枢和外周一些组织中NOSmRNA的含量 ,证明NOSmRNA广泛存在于中枢和外周组织中 ,在中枢以小脑含量为最高 ,在外周组织中以肾脏含量为最高 ;定量PCR方法测定NOSmRNA较Northernblotting分析更为灵敏Abstract: With Reverse Transcription Polymerase Chain Reaction(RT PCR),nitric oxide synthase(NOS) complementary DNA has been subcloned from the rat cerebellum and kidneys.The levels of NOS mRNA in some central and peripheral tissues in rats have been detected by Northern blotting and quantitative PCR.It has been proved that NOS mRNA existed extensively in central and peripheral tissues.NOS mRNA levels are highest in the cerebellum in the centre and in the kidney in peripheral tissues.Moreover,quantitative PCR is more sensitive than Northern blotting in detecting NOS gene expression.
