异搏定、尼卡地平对原发性高血压患者离体血管平滑肌细胞内钙的干预研究

A Study of Verapamil and Nicardipine on Cellular Calcium Metabolism of Cultured Human Arterial Smooth Muscle Cells in Essential Hypertensives

  • 摘要: 作者采用原发性高血压患者的离体动脉血管 ,并分离、培养动脉平滑肌细胞 ,观察了异搏定、尼卡地平对人动脉平滑肌细胞内钙的影响。由于原发性高血压患者动脉血管平滑肌细胞内钙的跨膜流动和细胞内释放机制尚不清楚 ,作者试图得到异搏定、尼卡地平对人离体动脉平滑肌细胞内钙释放和转运的直接证据。结果如下 :(1)原发性高血压患者动脉平滑肌细胞内总钙比正常血压组高 ,并与细胞内游离钙明显相关 ;(2 )两个钙通道拮抗剂均能显著减少两组的细胞内钙。异搏定显著减少4 5Ca2 +的跨膜内流 ,尼卡地平主要是阻止细胞内钙的释放

     

    Abstract: The experiment was performed on the isolated and cultured human arterial smooth muscle cells(HASMC) separated from the essential hypertensives.The effects of verapamil and nicardipine on the intracellular calcium of HASMC were investigated.The calcium transmembrane influx and intracellular releasing processes in ASMC of essential hypertensives have not been clearly defined,so the authors tried to evaluate and get the direct evidence of the effects of the above mentioned drugs on calcium transmitting and releasing in HASMC in vitro.The results are as follows:1.The total intracellular calcium of the essential hypertensives is much higher than that of the control group.And it has an obvious correlationship with the intracellular calcium cations.2.The two calcium channel blockers can significantly decrease the intracellular calcium in both groups.Verapamil has decreased the transmembrane influx of 45 Ca 2+ .Nicardipine mainly prevents the intracellular calcium releasing.
