Renal Generation of Dopamine from γ- L-Glutamyl-L-dopa is Impaired in Rabbits with Doxorubicin- induced Congestive Heart Failure

  • 摘要: 给正常家兔和阿霉素性心肌病心衰家兔静脉注射谷酰多巴 (2 5和 10 0μg/kg· min) 6 0 min后 ,平均动脉压、心率和血浆多巴胺含量均无显著变化。两组动物血浆谷酰多巴的浓度类似 ,尿多巴胺排泌率和肾组织多巴胺含量均显著增加 ,其增加程度在心力衰竭组显著低于正常对照组。表明阿霉素性心肌病心衰家兔肾脏由谷酰多巴合成多巴胺的功能受损

     

    Abstract: Normal saline,γ L glutamyl L dopa at 25 and 100 μg/kg·min were infused iv each for 60 min in control rabbits and rabbits with doxorubicin induced congestive heart failure.Mean arterial pressure,heart rate and plasma dopamine concentrations remained unchanged in both groups.Plasma L glutamyl dopa levels reached in the two groups were similar.Urine dopamine excretion and renal dopamine content were markedly increased after γ L glutamyl L dopa infusion.Moreover,γ L glutamyl L dopa was to a less degree endorenally converted into dopamine in the heart failure rabbits than in the controls.Therefore,rabbits with doxorubicin induced congestive heart failure exhibit a defective renal production of dopamine,which may contribute to renal vasoconstriction and water and sodium retention in heart failure.
