自发性高血压大鼠血管合成AngⅡ、醛固酮及醛固酮合成酶mRNA表达

Aldosterone Over-production and CYP11B2 mRNA Over-expression in Vessels of Spontaneously Hypertensive Rats

  • 摘要: 目的探讨血管合成血管紧张素Ⅱ及血管合成醛固酮在高血压大鼠中的变化。方法用肠系膜动脉离体灌注,在灌注液中测定血管紧张素Ⅱ及醛固酮。用RT-PCR及Southernblot法检测醛固酮合成酶基因表达的情况。结果SHR血管合成的血管紧张素Ⅱ及醛固酮增多、主动脉组织内醛固酮合成酶基因mRNA表达增强。结论SHR血管合成的血管紧张素Ⅱ及醛固酮增多,血醛固酮合成酶基因表达上调。

     

    Abstract: Aim\ To investigate the changement of AngⅡ, aldosterone and aldosterone synthetase in SHR.\ Methods\ In vitro the mesentery vessels were perfusion for determinated the AngⅡ(immumoassay method),aldosterone(HPLC method). Aldosterone synthetase(RTPCR and Southern blot method) in SHR Aorta.\ Results wre compared with Wistar rats.\ Results\ AngⅡ,aldosterone and its synthetase mRNA were elevated in the perfusion fluid and aorta tissue of SHR than the Wistar.\ Conclusion\ Aldosterone, as an important factor for hypertension, is demonstrated to be overproduced in mesemtry vasculature of SHR.\ The mRNA expression of aldosterone synthetase is up-regulated in aorta tissue of SHR.
