AngⅡ对乳鼠心肌细胞血小板衍生生长因子受体的调控作用

摘要: 目的 探讨血管紧张素Ⅱ (AngⅡ )对心肌细胞血小板衍生生长因子 β(PDGF β)受体含量的影响。 方法 分离纯化培养的乳鼠心肌细胞 ,以 10 -6molAngⅡ刺激作为实验 ,AngⅡ刺激前应用 10 -5mol洛沙坦 (AngⅡ的 1型受体特异性拮抗剂 )预处理作为拮抗组 ,以正常的乳鼠心肌细胞为对照组 ,免疫印迹法测定培养lh、6h、12h时心肌细胞PDGF β受体的含量。结果 AngⅡ刺激培养 1h、6h的乳鼠心肌细胞PDGF β受体显著上调 (P <0 0 5 ,<0 0 1) :洛沙坦完全了取消AngⅡ对PDGF β受体的上调作用。 结论 AngⅡ通过AT1受体诱导心肌细胞PDGF受体上调 ,此可能是AngⅡ致心肌细胞肥大的重要机制之一Abstract: Objective To investigate the effects of angiotension Ⅱ(AngⅡ) on the regulation of PDGF receptor level in cardiac myocytes. Methods The culturd neonatal rat cardiac myocytes were treated with 10 -6 mol Ang Ⅱ as experiment group and 10 -5 mol losartan (antagonist of AT 1 receptor) as antagonist group,with untreated neonatal rat cardiac myocytea as control group. The levels of PDGF β were detected by Western blot after treatment 1 h,6 h,12 h. Results\ Up regulation of PDGF β level by Ang Ⅱ at neonatal rat cardiac myocytes was found at 1 h( P <0 05) and 6 h( P <0 01). The increase in PDGF β level effect could be abolished by 10 -5 mol losartan.\ Conclusion The results indicate that the up regulation of PDGF β receptor levels by Ang Ⅱ were mainly mediated by AT 1 receptor.\ This may be a important mechanism for myocardium hypertrophy.
