不同剂量卡托普利对动脉粥样硬化影响的实验研究

Effect of Different dose Captopril on Atherosclerosis in Rabbits

  • 摘要: 目的 探讨不同剂量卡托普利对动脉粥样硬化的影响。方法 将纯种日本大耳白兔随机分为对照组、胆固醇组、卡托普利高剂量组、中剂量组及低剂量组。建立家兔动脉粥样硬化模型观察各组家兔血脂水平及主动脉的病理学改变 ;采用原位逆转录PCR法测定各组原癌基因c myc和c fosmRNA的表达水平。结果  (1)与胆固醇组比较 ,卡托普利各剂量组的血脂水平无显著性差异 ,但病理形态学发现 ,高剂量组的主动脉斑块面积比明显降低 (P <0 0 5 ,P <0 0 1) ,泡沫细胞层明显减少 ,低剂量组与胆固醇组比较无显著性差异。 (2 )胆固醇组的原癌基因c myc、c fosmRNA表达的阳性率较对照组显著增高 ,而高剂量组的阳性率则显著低于胆固醇组 (P <0 0 5 ) ,中、低剂量组与胆固醇组比较无显著差异。结论 只有用高剂量的卡托普利才能有效地防止动脉粥样硬化斑块的形成及阻止动脉粥样硬化的发展。

     

    Abstract: Aim To investigate the effect of different doses of captopril on atherosclerosis in rabbits. Methods 53 male Japanese white rabbits were randomly divided into control group, cholesterol group and captopril group of high , middle , low dose. The change of serum lipid concentrations and the histopathological changes of aorta were studied. The positive expression of pro oncogene c myc and c fos mRNA was also studied with in situ reverse transcription PCR (ISRT PCR). Results (1)Serum lipid levels in captopril group had no significant change compared with cholesterol group. The ratio of aorta plaque areas in high dose captopril group was significantly lower than that of the cholesterol group ( P <0 01) with the foam cells being remarkably decreased. No significant difference was observed between of low dose captopril and cholesterol group. (2) Positive expression rate of pro oncogene c myc and c fos mRNA was significantly higher in cholesterol group than in the control group. The rate of positive expression was remarkably lower in the high dose captopril group than in the cholesterol group ( P <0 05). No significant difference was found between groups in middle,low dose groups or in the cholesterol group. Conclusions The study had suggested that inhibits the formation of aorta plaque and prevent atherosclerotic development,a higher dose of captopril is likely necessary.
