血管紧张素(1~7)对鼠系膜细胞和OK细胞增殖的影响
Effects of Angiotensin(1~7) on the Proliferation of Cultured SHR Glomerular Mesangial Cells and Opossum Kidney Cells

摘要: 目的 观察血管紧张素 (1~ 7) Ang(1~ 7) 对自发性高血压大鼠 (SHR)肾小球系膜细胞和负鼠肾小管OK细胞 (OK细胞 )增殖的影响。方法 采用体外培养的SHR肾小球系膜细胞和OK细胞 ,通过3 H 胸腺嘧啶核苷掺入法观察这两种细胞对血管紧张素Ⅱ (AngⅡ )及Ang(1~ 7)增殖变化。结果 (1)AngⅡ对SHR肾小球系膜细胞和负鼠肾小管OK细胞DNA合成的刺激作用 ,随着浓度的升高 (10 -10 ~ 10 -6mol/L)作用加强 ,且呈剂量依赖关系 ,而浓度进一步升高到 10 -5mol/L时 ,作用反而减弱。 (2 )当培养液中同时加入AngⅡ和Ang(1~ 7)时 ,随着Ang(1~ 7)浓度的升高 ,抗AngⅡ增殖作用逐步增强。 (3)当培养液中同时加入AngⅡ和血管紧张素转换酶抑制剂苯那普利拉 (Benazepri lat)时 ,随着Benazeprilat浓度的升高 ,抗AngⅡ增殖作用逐步增强。 结论 (1)AngⅡ能刺激自发性高血压大鼠肾小球系膜细胞和负鼠肾小管OK细胞增殖 ;(2 )Ang(1~ 7)和Benazeprilat能抑制AngⅡ引起的细胞增殖作用 ,对肾脏细胞可能有直接保护作用 ;(3)它们的抑制作用呈剂量依赖关系。Abstract: Aim Angiotensin(1~7)has been shown to inhibit the proliferation of smooth vascular cells, while the effect of whom on SHR glomerular mesangial cell(GMC) and opossum kidney cells(OK) are not clear.\ We determined to study the effect of Ang(1~7)on these two kind of cells. Methods The proliferation of cultured glomerular cells and OK cells were detected by measuring 3H thymidine incorporation. Results The synthesis of DNA in cultured glomerular mesangial cells of SHR and opossum kidney cells were stimulated by Ang Ⅱ in the range of 10 -5 ~10 -6 mol/L, however the effect was blunted when the concentration reached 10 -5 moL.\ Ang(1~7) in the range of 10 -10 ~10 -6 mol/L dose\|dependently inhibited the proliferative effect induced by Ang Ⅱ(10 -6 mol/L) in GMC and OK.\ Benazeprilat, a metabolic product of benazepril, demonstrated a potential antagonist effect on the proliferation of GMC and OK cells by AngⅡ. Conclusion (1)Ang Ⅱ stimulated the growth of cultured glomerular mesangial cells of spontaneously hypertensive rats and opossum kidney cells; (2)Ang(1~7) and benazeprilat inhibite the proliferation induced by angiotensin Ⅱ in cultured glomerular mesangial cells of SHR and OK cells.\ Angiotensin(1~7) and Benazeprilat may have renal protective effect.
