The effect of Norepinephrine on Apoptosis and Hypertrophy of Cardiac Myocytes

  • 摘要: 目的 探讨去甲肾上腺素 (NE)对心肌细胞凋亡、肥大的影响。方法 培养乳鼠心肌细胞暴露于三种浓度NE(2、2 0、2 0 0 μmol/L) 36小时 ,检测心肌细胞凋亡率、DNA梯状带纹、心肌细胞蛋白含量、3 H -亮氨酸掺入率的变化。结果高、中两种浓度NE可导致心肌细胞凋亡率明显增高和DNA梯状带纹出现 ,高、中、低三种浓度NE均能明显增加心肌细胞蛋白含量和3 H -亮氨酸掺入率。结论 NE能诱导培养幼鼠心肌细胞凋亡和肥大。高、中、低浓度的NE ,均可促进心肌细胞肥大 ,而较高浓度的NE方诱导心肌细胞凋亡。

     

    Abstract: Objective To study the effects of norepinephrine on apoptosis and hypertrophy of cardiac myocytes. Methods The cultured neonatal rat cardiomyocytes (NRCs) were exposed to NE (2 μmol/L,20 μmol/L,200 μmol/L) in free-serum medium for 36 h. The percentage of apoptotic myocytes,DNA ladder, 3H-Leucine incorporation and cellular protein content were investigated. Results NE at the concentration of 20 μmol/L and 200 μmol/L could increased the apoptotic cells. NE-induced increases in 3H-Leucine incorporation and cellular protein content at all concentration. Conclusion Both low- and high-concentration of NE induced hypertrophy in cultured NRCs y,while only high but not low concentration NE caused apoptosis.
