糖尿病大鼠心钠素水平的动态观察及依那普利的调节作用

Gene Expression of Atrial Natriuretic Peptide in Ventricles and Plasma and the Effect of Enalapril in Diabetic Rats

  • 摘要: 目的观察糖尿病大鼠心钠素心肌及循环水平的动态变化及依那普利对其表达的影响。方法成年雄性Wistar大鼠分为:1月对照组和糖尿病组(各7只);3月对照组和糖尿病组(各7只);5月对照组、糖尿病组和依那普利治疗组(各8只)。分别于病程1、3、5月经心脏采血,放免法测定血浆ANP水平;记录心脏重量指数;采用半定量RT-PCR分析1、5月程左心室ANPmRNA水平。结果糖尿病大鼠血浆ANP水平(pg/mL)于病程1月(1528.5±354.2vs1134.1±368.3,P=0.064)及3月(1747.8±346.2vs1220.1±355.1)pg/mL,(P<0.05)明显升高,5月(1047.9±317.4vs914.1±205.0)pg/mL,(P=ns)与对照组无差异,且显著低于3月组(P<0.05);其左室ANPmRNA含量与对照组比较,病程1月(1.559±0.501vs1.041±0.404)pg/mL,(P=0.051)升高,5月(1.317±0.362vs1.177±0.472)pg/mL,(P=ns)无差异。依那普利治疗组大鼠左室ANP表达无明显增强,但血浆ANP浓度显著高于未治疗组(1405.9±360.7vs1047.9±317.4)pg/mL,(P<0.05)及对照组(1405.9±360.7vs914.1±205.0)pg/mL,(P<0.05)。结论糖尿病大鼠ANP心脏表达及循环水平呈现先升高后降低的变化趋势;依那普利干预明显提高糖尿病大鼠血浆ANP水平,可能是其发挥抗心衰及心脏保护作用的机制之一。

     

    Abstract: Objective To observe the gene expression of atrial natriuretic peptide (ANP) in ventricles and plasma and the effect of enalapril treatment in diabetic rats by streptozicin 60 mg/kg I.P.Methods Plasma ANP concentrations were determined in Wistar rats with RIA at 1, 3, 5 month after establishment of experimental diabets model. Semi-quantitative RT-PCR was used to evaluate the left ventricular ANP mRNA content, and heart weight index(HWI) were recorded. A group of diabetic rats were treated with enalapril(2 mg/kg·d) for 5 months.Results The plasma ANP levels in diabetic rats were increased at 1-month (1528.5±354.2 vs 1134.1±368.3)pg/mL(P=0.064) and at 3-month (1747.8±346.2 vs 1220.1±355.1)pg/mL(P<0.05) as compared to control. At 5-month, however, the plasma concentrations of ANP were not different between diabetics and control(1047.9±317.4 vs 914.1±205.0)pg/mL(P=NS). The left ventricular ANP mRNA contents in diabetic rats were increased at 1-month (1.559±0.501 vs 1.041±0.404)pg/mL(P=0.051) and then were reversed to normal level at 5-month (1.317±0.362 vs 1.177±0.472)pg/mL(P=NS). In enalapril-treated diabetic rats, the plasma ANP concentrations were significantly higher than that in diabetic rats(1405.9±360.7 vs 1047.9±317.4)(pg/mL)(P<0.05) and control (1405.9±360.7 vs 914.1±205.0)pg/mL(P<0.05), while the gene expression were not augmented. Conclusion Enhanced ventricular gene expression and elevated plasma levels of ANP was found at the early stage of diabetes.Enalapril treatment increased the plasma ANP concentrationsin diabetic rats, which is perhaps one of mechanisms underlying the effect of enalapril on heart function improvement and myocardium preservation.
