原发性高血压患者动脉血管组织E26转录因子1及单核细胞趋化蛋白1的表达和临床意义
Expression and clinical implications of E26 transformation-specific-1 and monocyte chemoattractant protein-1 in arterial tissues of essential hypertensives

摘要: 目的观察E26转录因子1(Ets-1)及其下游靶基因的表达产物单核细胞趋化蛋白1(MCP-1)在原发性高血压患者动脉血管组织中的表达,探讨其在原发性高血压患者血管重构中的作用。方法收集2010-07-12安徽医科大学第三附属医院(合肥市第一人民医院)原发性高血压患者(高血压组,n=17)及正常血压者(正常血压组,n=16)手术废弃组织中的动脉血管,采用免疫组织化学法结合图像信号分析技术及逆转录聚合酶链反应(RT-PCR)方法,检测动脉血管组织中Ets-1、MCP-1蛋白及mRNA的表达。结果高血压组动脉血管组织中Ets-1肠系膜动脉(186.4±2.4)比(102.8±1.9);肋间动脉(153.4±2.0)比(101.2±1.6)、MCP-1肠系膜动脉(115.3±1.0)比(83.2±0.7);肋间动脉(122.4±0.8)比(70.1±0.6)蛋白及mRNA的表达明显高于正常血压组(均P<0.05);Pearson相关分析显示,高血压组动脉血管组织Ets-1与MCP-1mRNA表达呈正相关(肠系膜动脉,r=0.87;肋间动脉,r=0.83,均P<0.01)。结论与正常血压者相比,Ets-1和MCP-1在原发性高血压患者血管组织中的表达明显上调,两者的表达呈正相关。提示Ets-1、MCP-1在原发性高血压血管重构可能起重要作用。Abstract: Objective To investigate the expression of E26transformation-specific-1(Ets-1)and its downstream target gene expression product monocyte chemoattractant protein-1(MCP-1)in arterial tissues,and to explore their roles in vascular remolding of essential hypertensive patients. Methods From July 2010to December 2010,samples of arteries from hypertensive patients(hypertensive group,n=17)and normotensive controls(normotensive group,n=16)after surgeries were collected in Third Affiliated Hospital of Anhui Medical University(First People’s Hospital of Hefei). The protein and mRNA levels of Ets-1and MCP-1in artery tissues were detected by immunohistochemical method and reverse transcription-polymerase chain reaction(RT-PCR). Results As compared with the normotensive group,the protein expressions of Ets-1and MCP-1in hypertensive mesenteric arteries(186.4±2.4)vs(102.8±1.9);(115.3±1.0)vs(83.2±0.7)and intercostals arteries(153.4±2.0)vs(101.2±1.6);(122.4±0.8)vs(70.1±0.6),all P<0.05increased markedly. So did the mRNA levels of Ets-1and MCP-1.A positive correlation between Ets-1and MCP-1in arterial tissues of hypertensive patients was found by Pearson correlation analysis with r=0.87in mesenteric arteries and r=0.83in intercostal arteries(all P <0.01).Conclusions Compared with the normotensive group,the expressions of Ets-1and MCP-1are significantly up-regulated in vessel tissues of hypertensive patients. Furthermore,the expression of Ets-1and MCP-1is positively correlated.Which implicates that Ets-1,MCP-1may play an important role in vascular remolding of essential hypertension.
