熊胆对自发性高血压大鼠肠系膜动脉血管结构和舒张功能的影响

Effect of Xiongdan on mesenteric vascular structure and dilatation function in spontaneously hypertensive rats

  • 摘要: 目的观察熊胆对自发性高血压大鼠(SHR)血压、肠系膜动脉血管结构和舒张功能的影响。方法 12周龄雄性SHR 16只随机分为:SHR熊胆治疗组SHR-X,n=8,800mg/(kg·d)和SHR对照组(SHR,n=8),并选取对应周龄雄性Wistar Kyoto大鼠作为正常血压对照组(WKY,n=8),灌胃8周。采用无创尾袖法测量大鼠尾动脉收缩压。应用计算机图像分析系统计算HE染色后的血管管壁面积/管腔面积(W/L)、管壁厚度/管腔半径(WT/LR)。采用PowerLab生物信息采集系统检测离体肠系膜动脉对不同浓度血管活性物质的舒张反应。结果熊胆治疗4和8周末,SHR-X组的血压明显低于未治疗的SHR组收缩压,4周:SHR-X(176.4±11.4)比SHR(200.3±13.9)mm Hg;8周:SHR-X(169.4±12.0)比SHR(189.9±10.1)mm Hg;均P<0.01。熊胆治疗8周末,SHR-X组的肠系膜动脉W/L及WT/LR明显低于未治疗的SHR(W/L:SHR-X 0.53±0.09比SHR 1.82±0.96;WT/LR:SHR-X 0.24±0.08比SHR 0.53±0.29,均P<0.01),与WKY组比较差异无统计学意义(P>0.05);SHR-X的肠系膜动脉内皮依赖性血管舒张功能明显高于未治疗SHR最大舒张率:SHR-X(59.29±15.15)%比SHR(20.69±6.31)%,舒张反应敏感性(pD2):SHR-X 8.24±0.32比SHR 5.82±0.23;均P<0.01,非内皮依赖性血管舒张功能也明显高于SHR组最大舒张率:SHR-X(96.37±1.87)%比SHR(29.04±4.56)%;pD2:SHR-X 7.79±0.15比SHR 5.31±0.14;均P<0.01。结论熊胆治疗可轻度降低SHR的血压,明显改善肠系膜动脉血管结构和舒张功能。

     

    Abstract: Objective To observe the effect of Xiongdan on blood pressure,vascular structure and dilatation function of mesenteric arteries of spontaneously hypertensive rat(SHR). Methods Sixteen 12weeks-old male SHR were randomly divided into 2 groups:Xiongdan treated group SHR-X,n=8,800 mg/(kg·d)and untreated group(SHR,n=8),with age and weight matched Wistar Kyoto(WKY)rats served as control group(WKY,n=8). All rats were gavaged for 8weeks. Systolic blood pressure was measured by tail-cuff method before and after 4 and 8 weeks of treatment,respectively. The wall-to-lumen area ratios(W/L)and wall thickness-to-lumen radius ratios(WT/LR)of HE stained mesenteric arteries was assessed morphometrically by computer. The vasoactive substance-induced dilation reactivity of mesenteric arteries in vitro were measured at different concentrations by PowerLab biological signal analytical system. Results After 4 and 8 weeks of Xiongdan treatment,systolic blood pressure of SHR-X group was significantly lower than SHR group(176.4±11.4)vs(200.3±13.9)mm Hg at the 4th week;(169.4±12.0)vs(189.9±10.1)mm Hg at the 8th week;both P<0.01. At the end of the 8th week,W/L and WT/LR of mesenteric arteries of SHR-X group were markedly lower than SHR group(0.53±0.09vs1.82±0.96 and 0.24±0.08 vs 0.53±0.29,respectively,both P<0.01),with no statistic difference compared to WKY group(P >0.05). Compared to SHR group,both endothelium dependent relaxation Emax(59.29±15.15)% vs(20.69±6.31)%,pD2(8.24±0.32)vs(5.82±0.23),respectively,both P<0.01and endothelium independent relaxationEmax(96.37±1.87)% vs(29.04±4.56)%;pD2(7.79±0.15)vs(5.31±0.14),respectively;both P <0.01of mesenteric arteries were increased in SHR-X group after 8 weeks of treatment.Conclusion The Xiongdan treatment may mildly lower the blood pressure,and ameliorate the vascular structure and dilatation function of mesenteric arteries of SHR.
