过氧化物酶体增殖物激活受体的多个单核苷酸多态性与平均动脉压的关系

Association of the single nucleotide polymorphisms of peroxisome proliferator-activated receptor genes with mean arterial pressure

  • 摘要: 目的探讨过氧化物酶体增殖物激活受体(PPAR)α/δ/γ基因10个单核苷酸多态性(SNP)与平均动脉压(MAP)之间的关系。方法从江苏省代谢综合征和多代谢异常综合防治队列研究中,采用单纯随机抽样的方法抽取研究对象820例,男性270例,女性550例。根据最小等位基因频率≥5%、与多代谢异常有关联和位于基因片段功能区或可能改变功能区域3个标准,选取用于本研究的3个PPARα、2个PPARδ和5个PPARγ的SNP位点并进行检测,并分析PPAR基因10个SNP与MAP的关联。结果对选取的PPARα/δ/γ三亚型的10个SNP进行Hardy-Weinberg平衡检验分析显示,比较频数期望值和实际值发现,本研究选择的SNP位点的基因型差异无统计学意义。PPAR的SNP与MAP的关联分析结果表明,与野生型携带者相比,PPARα的rs135539突变基因型(A/C-C/C)和PPARδ的rs9794突变基因型(C/G-G/G)携带者的MAP分别变化-2.212(95%CI-3.715-0.711)和-1.580(95%CI-3.121-0.068)mm Hg,而PPARγ的rs10865710的突变基因型(C/G-G/G)携带者的MAP水平升高1.601(95%CI 1.018~3.021)mm Hg,均P<0.05。结论 PPARα的rs135539、PPARδ的rs9794与PPARγ的rs10865710 3个SNP与MAP相关。

     

    Abstract: Objective To investigate the relationships between ten single nucleotide polymorphisms(SNP)of peroxisome proliferators-activated receptor(PPAR)α/δ/γgenes and mean arterial pressure(MAP). Methods A total of 820 subjects from "the prevention of metabolic syndrome and multi-metabolic disorders in Jiangsu province of China study(PMMJS)" were included,which was composed of 550 females and 270 males. Ten SNP of PPARα/δ/γgenes were selected according to the following criteria:(1)minor allele frequency(MAF)≥0.05;(2)reported the association with metabolic syndrome,and(3)locus at functional region or possible occurrence of mutation lying in the area. The association between SNP and MAP was analyzed. Results Hardy-Weinberg equilibrium analysis was used to evaluate ten SNP of PPARα/δ/γgenes,showing that all of selected SNP satisfied Hardy-Weinberg equilibrium(P>0.05). Multiple linear regression analysis showed that compared to the persons with wild genotype of PPAR genes,the MAP levels in subjects with mutant genotype of rs135539 in PPARα(A/C-C/C)and rs9794 in PPARδ(C/G-G/G)decreased 2.212(95% CI-3.715 to-0.711)mm Hg and 1.580(95% CI-3.121 to-0.068)mm Hg,respectively. However,the mean level of MAP in the subjects with mutant genotype of rs10865710 in PPARγgenes(C/G-G/G)increased 1.601(95% CI 1.018 to 3.021)mm Hg,all P <0.05.Conclusion rs135539 of PPARαgene,rs9794 of PPARδgene and rs10865710 of PPARγgene are significantly associated with MAP levels.
