基于磁共振技术的应激性高血压大鼠模型血清代谢组学研究

The study of serum metabolic changes based on nuclear magnetic resonance imaging in stress induced hypertensive rats

  • 摘要: 目的建立应激性高血压(SIH)模型大鼠,探讨其血清代谢组分的变化。方法选取鼠龄在6~8周的Wistar大鼠(雄性)30只,随机分为SIH模型组(20只)与对照组(10只),适应性喂养7d后,SIH模型组每天制动、夹尾、间断足底电刺激、声光电刺激等进行SIH模型的构建,持续刺激9周后,SIH模型组最终造模成功6只,对照组同期进行正常喂养。通过采用磁共振(NMR)技术结合模型识别和正交偏最小二乘判别(OPLS-DA)分析方法检测SIH模型大鼠血清中代谢组分的变化。结果与对照组相比,SIH模型组大鼠血清中糖蛋白、β-葡萄糖、极低密度脂蛋白(VLDL)、乙酰乙酸、肉碱、异亮氨酸、亮氨酸、缬氨酸含量升高(相关系数r分别为-0.71、-0.76、-0.80、-0.75、-0.82、-0.73、-0.82、-0.72;均P<0.05)。结论 SIH模型大鼠存在脂代谢、糖代谢和氨基酸代谢异常。

     

    Abstract: Objective To study the changes of serum metabolic components in stress induced hypertension(SIH)rats. Methods Thirty male Wistar rats aged 6-8 weeks were randomly divided into SIH model group(n=20)and normal control group(n=10). After 7 days of adaptive feeding,SIH model group was established through daily braked,tail clamp,intermittent plantar stimulation,sound and light stimulation. After 9 weeks of continuous stimulation,the SIH model was successful established on 6 rats. Rats in control group were given normal feeding during the same period. The changes of metabolic components in serum of SIH model rats were detected by nuclear magnetic resonance(NMR)technique combined with model identification and orthogonal partial least squares(OPLS-DA). Results Compared with the control group,the levels of glycoprotein,β-glucose,very low denisity lipoprotein(VLDL),acetoacetic acid,carnitine,isoleucine,leucine and valine in the serum of SIH model group were significantly higher(r=-0.71,-0.76,-0.80,-0.75,-0.82,-0.73,-0.82,-0.72,respectively;all P<0.05). Conclusion Lipid,glucose and amino acid metabolisms in SIH rats were significantly different from those in control rats.
