The Expression of Extracellular Signal-regulated Kinsae -2 (ERK-2) and c-jun in Renal Vascular Smooth Muscle Cell in Hypertensive Rats

Abstract
Objective To explore the expression of extracellular regulated kinase (ERK-2) and protoncogene c-jun in vascular smooth muscle cell (VSMC) in hypertension. Methods Two kidney-one clip hypertension model was established in Wistars rats. The expression of ERK-2 and c-jun proto-oncogene was studied using immunohistochemical technique. Results SBP was raised significantly in hypertension rats from the baseline of (112±18) mmHg to (198±33) mmHg at the end of fourth week. Glomerular fibrosis in hypertensions rats were significantly greater than that of the control. Hyaline degeneration and thickening arterial wall with profound by increases in VSMC in renal arteries. Incidence rate of positive ERK-2 in VSMC in efferent, interlobular, interlobar and arch arteries was higher in hypertension rats than that in control(7.15 vs 3.0, 14.6 vs 6.7, 13.6 vs 11.1, 29.4 vs 19.9. P<0.05 or 0.01). The c-jun staining positive rate of VSMC in efferent, interlobular, and arch arteries was higher in hypertension than that of controls (8.7 vs 3.1, 12.4 vs 6.8, 21.1vs 5.9. P<0.01). Conclusion In two kidney-one clip hypertensive rats, overexpression of ERK-2 and activated c-jun were associated with the VSMC proliferation in renal arteries.
